For my project I want to find the origin of replication on a megaplasmid I am working with.  I will do this by using GC skew and scanning motifs in that region.  This will then allow me to create smaller plasmids containing the origin of replication to test for host specificity of the plasmid among Pseudomonads.  Here is a concept map of the project.
